چکیده

The present study is concerned with the production of cephalosporin C by Cephalosporium acremonium C-10. Two culture media were tested for the production of antibiotic. Among these culture media M1 containing sucrose, ammonium acetate, cornstarch, sugar cane molasses, soybean meal, calcium carbonate, calcium sulphate and methyl oleate gave optimum production of cephalosporin C. The cultural conditions such as pH 6.5 and temperature 30°C were optimized. The production of antibiotic was found optimal 48 h after conidial inoculation. The effect of size of inoculum was studied and the best results were obtained at 1ml of inoculum size.

Synthesis of cephalosporin C from sulfate by mutants of Cephalosporium acremonium.

The innate ability of Cephalosporium acremonium to use methionine preferentially over sulfate for synthesis of cephalosporin C can be influenced through mutation. Mutants of C. acremonium with altered capacity to utilize sulfate for synthesis of antibiotic were isolated and partially characterized with respect to the uptake of sulfate and the regulation of arylsulfatase.

Production of cephalosporin C by single and double sulfur auxotrophic mutants of Cephalosporium acremonium.

An early blocked sulfur amino acid auxotroph, Cephalosporium acremonium mutant 274-1 (which could be satisfied by methionine or cysteine), utilized organic sulfur compounds for cephalosporin C production in the following order of decreasing effectiveness; methionine > cystathionine > cysteine, despite the fact that cysteine is considered to be the immediate precursor of the antibiotic. When a g...

Changes in Cellular Fatty Acid Composition of Cephalosporium acremonium during Cephalosporin C Production.

Cephalosporium acremonium was cultivated in fermentation medium containing sucrose or methyl oleate as a carbon source for cephalosporin C production. The level of antibiotic production was 48 g of cephalosporin C per liter under optimum conditions when methyl oleate was used. The C(18:1) (oleic acid) methyl ester appeared to be utilized faster than the C(18:2) (linoleic acid) methyl ester in f...

Process strategies for cephalosporin C fermentation

Cephalosporin C (CPC), a β-lactam antibiotic, is starting molecule for industrial production of semi synthetic cephalosporins. CPC fermentation has been carried out in Air Lift Reactor (indigenously made up of borosilicate glass) in different fermentation modes i.e., batch and fed batch modes using a mold Acremonium chrysogenum (synn. Cephalosporium acremonium). A definite correlation exists be...

Incorporation of acetate into cephalosporin C.

Cephalosporin C, an antibiotic with structure (I) (Abraham & Newton, 1961; Hodgkin & Maslen, 1961), was first isolated from an impure preparation of the chemically related penicillin NJ [(D-4amino-4-carboxybutyl)penicillin] that had been obtained from the culture fluid of a Cephalosporium sp. (Newton & Abraham, 1956; Abraham & Newton, 1956). The amount of cephalosporin C produced by this organi...
